TREM2 Effector Reporter Cell

I. Background

      TREM2是免疫球蛋白超家族的單次跨膜受體，主要表達(dá)于巨噬細(xì)胞、未成熟單核細(xì)胞 衍生的樹突狀細(xì)胞、破骨細(xì)胞和小膠質(zhì)細(xì)胞上。TREM2 包含免疫球蛋白 V 型結(jié)構(gòu)的胞外 域、跨膜域和沒有信號(hào)傳導(dǎo)基序的短胞內(nèi)結(jié)構(gòu)域。它通過 DNAX 激活蛋白 10 和 12 （DAP10 和 DAP12）發(fā)出信號(hào)。單個(gè) TREM2 分子可以同時(shí)與 DAP10 和 DAP12 相互作 用。

      TREM2 與陰離子配體結(jié)合，例如磷脂、硫苷脂、細(xì)菌脂多糖(LPS) 和 DNA。TREM2– DAP12–DAP10信號(hào)傳導(dǎo)觸發(fā)蛋白質(zhì)和脂質(zhì)磷酸化級(jí)聯(lián)，導(dǎo)致 Ca2+ 動(dòng)員、整合素激活、細(xì)胞骨架重排、雷帕霉素的機(jī)制靶點(diǎn)(mTOR) 和絲裂原活化蛋白激酶(MAPK) 信號(hào)傳導(dǎo)以及能 量代謝的激活。

 
II. Description

      TREM2 Effector Reporter 報(bào)告基因藥靶模型很好的模擬了體內(nèi) TREM2 的信號(hào)轉(zhuǎn)導(dǎo)過程， 原理見下圖所示。
